Epistasis and tradeoffs in ultra-high-throughput mutational scans
We are developing a platform for characterizing millions of bacterial strains using microfluidics, high-throughput fluorescent microscopy, and a petabyte-scale data analysis pipeline. We are applying this platform to observe epistasis and tradeoffs between multiple phenotypes in fluorescent proteins.
Predictibility of sigma-antisigma sequestration circuits
We are performing detailed quantitative measurements and modeling of sequestration-based positive- and negative-feedback circuits built with sigma-antisigma factor pairs.
Co-translational folding and other determinants of codon choice
We are applying our fluorescent protein screening platform to measure misfolded protein stress as a function of codon choice.
